EFEKTIFITAS SEDUHAN DAUN KERSEN (Muntingia calabura L.) TERHADAP KADAR SGOT & SGPT PADA TIKUS DIABETES MELITUS YANG DIINDUKSI STREPTOZOCIN-NICOTINAMIDE (STZ-NA)
Abstract
Diabetes mellitus is a degenerative disease that caused many complications where in diabetes mellitus blood glucose levels is increase and oxidative stress. Oxidative Stress and high blood glucose levels can cause liver damage and characterized by the increase in liver enzymes are AST and ALT. Cherry (Muntingia calabura L.) is one kind of plant that is easy to live in Indonesia and thrives around the neighborhood. The previous study mentions that cherry has antioxidant activity because it contains flavonoids. These antioxidants can be used to prevent oxidative stress. The purpose of this study was to examine the effectiveness of cherry leaves steeping in decreased levels of SGOT and SGPT rat (Rattus novergicus) diabetes mellitus induced by Streptozotocin-nicotinamide (STZ-NA).
Methods: This study is an experimental research study design with pre and post test control design. The subjects are 30 white rats Sprague Dawley were divided into 5 groups: group 1 (negative control), group 2 (metformin), group 3 (steeping leaves of cherry 250 mg/200 grBW), group 4 (steeping leaves of cherry 500 mg/200 grBW), and group 5 (cherry leaves steeping 750 mg/200 grBW). The duration of the study was 26 days. 1-5 group induced with streptozotocin dose of 65 mg/kgBW and nicotinamide 230 mg/kgBW for 5 days until the rats be diabetes mellitus (fasting blood sugar >135 mg/dl) and then given treatment for 14 days. Cherry leaves steeping made by mixing dried cherry leaves with water and suitable dosage is given according to the weight of each rat. Intake levels of GDP using GOD-PAP enzymatic method, whereas SGOT and SGPT using a UV-Vis spectrophotometer. Data were analyzed using paired t test and One Way Anova.
Results: The results of statistical tests with paired t-test showed significant differences in the levels of SGOT and SGPT before and after treatment (p =0.0001). One way Anova test on average there are distinct decrease in each group (p =0.0001). Steeping most effectively reduce levels of SGOT and SGPT ie the dose 750 mg/200 grBW. Conclusion: From this study it can be concluded that the administration cherry leaves steeping can reduce levels of SGOT and SGPT with the optimal dose of 750 mg/200 grBW but further research is needed to a maximum dose that SGOT sample to be normal.
