UJI AKTIVITAS ANTIINFLAMASI ISOLAT ALKALOID LADA (Piper nigrum L.) PADA TIKUS GALUR WISTAR: STUDI IN VIVO DAN IN SILICO
Abstract
Inflammation occurs as the attempt of body to inactivate organisms that attack the body, removing irritants and regulate tissue repair. One of the medicinal plants used empirically as antiinflammation is the pepper (Piper nigrum L.).
This study aims to analyze the anti-inflammatory activity of isolate alkaloid of pepper (Piper nigrum L.) against to cyclooxygenase enzyme in Wistar rats that induced by carrageenin in accordance with in vivo and in silico test used molecular docking method. This study used experimental design, the strain wistar male rats as test animals. As much as 25 rats, devided into 5 groups. Group I: without treatment. Group II: Natrium Diclofenac as comparative compound dose of 13.5 mg / kgBB. Group III. IV, V: use isolate alkaloid of pepper dose of 5; 10 and 15 mg/kgBB. The measurement result of udem volume is measure the value of Area Under Curve (AUC) and % of anti-inflammatory power then the data is analyzed to understand the differences among groups. Besides in vivo test, in silico test also apply in this research using molecular docking method with auto dock tools to the COX-2 as target receptor.
The result shows, the provision of isolate alkaloid of pepper (Piper nigrum L.) dose of 15 mg/KgBB to rats will increase the Anti-Inflammatory Power (50, 59%) in foot of edema wistar rats model that induced by carrageenin. Based on the results of Tukey HSD test with a level of 95% shows that isolate alkaloid of pepper dose of 15 mg/kgBB is not significantly different from the comparative compound natrium diclofenac dose of 13.5 mg/kgBB. Isolate alkaloid of pepper dose of 15 mg/kgBB has ability to inhibit inflammatory as well as natrium diclofenac dose 13.5 mg / kgBB. In silico test of some marker compound pepper (Piper nigrum) that have potential to be developed as an anti-inflammatory agent, Piperine with the value of binding energy of -8.0 kcal/mol is more powerful than the comparative compound natrium diclofenac (docking score: -6.9 kcal / mol). The results of visualization showed that the test compound piperine and comparative compound were bonded with the same residue, that was leucine 352, valine 349, valine 523, and alanine 527. The conclusion of this study is alcaloid isolate from Piper nigrum have activity as an anti-inflammatory agents suspected to inhibit the cyclooxygenase enzyme.
